Abstract

Indole-3-acetic acid (IAA), a key phytohormone widely synthesized by soil bacteria, plays a vital role in root development and stress resilience. This study aimed to optimize the culture conditions for IAA production by Kocuria rosea SS1 and to evaluate the efficacy of bacterial derived IAA on the micropropagation of potato (Solanum tuberosum L. cv. Spunta). Maximum IAA production (10.83 mg 100 mL− 1, as quantified by HPLC) was achieved in nutrient broth at pH 7 and 30 °C, supplemented with 1 mM tryptophan and incubated under shaking conditions for three days. Filter-sterilized bacterial supernatant containing IAA (0.0–1.0 mgL− 1) was incorporated into Murashige and Skoog (MS) medium and compared to commercial IAA in potato tissue culture. After 28 days, plantlets treated with bacterial IAA, particularly at 0.25 mgL− 1; showed significant enhancements in fresh weight (1.5-fold), roots number (69.9%), leaves number (100%) and shoot length (50%) compared to the control, outperforming equivalent doses of synthetic IAA. Biochemical analyses revealed increased levels of total sugars, chlorophylls, carotenoids, and endogenous IAA and gibberellins. These findings demonstrate that the bacterial supernatant serves as a potent, eco-friendly biostimulant that enhances the growth and biochemical quality of in vitro-propagated potato plantlets, offering a sustainable alternative to synthetic growth regulators.

Indole-3-Acetic Acid Production by Klebsiella variicola from Watermeal and Its Role in Enhancing Watermeal Growth

Plant growth-promoting bacteria (PGPB) enhance plant development through mechanisms such as the production of indole-3-acetic acid (IAA). However, endophytic bacteria associated with aquatic plants, particularly watermeal (Wolffia spp.), remain poorly explored. This study investigated the potential of bacterially deriv...

Enhancing the Resistance of Soybean Plants to Waterlogging Stress Using Gamma-Irradiated Priestia megaterium

Results indicate that gamma-irradiated P. megaterium mutants can differentially modify soybean root system responses to flooding stress, supporting gamma-ray irradiation as a useful approach for developing PGPR with improved functions for agricultural applications under waterlogging stresses.

Effects of Plant Growth Regulators on Micropropagation and Silymarin Accumulation in Milk Thistle (Silybum marianum L.) In Vitro

Milk thistle (Silybum marianum L.) is a valuable medicinal plant known for its hepatoprotective compound, silymarin. However, the silymarin content varies with genotype, cultivation methods, climatic conditions, and harvest time. Micropropagation provides a solution by producing genetically uniform plantlets and offeri...

Biostimulant potential of lactic acid bacteria in hydroponic lettuce production: growth and nutritional quality

The use of microbial biostimulants has become an important strategy for improving the productivity and quality of soilless vegetable production systems. However, the potential role of lactic acid bacteria (LAB) in hydroponic crop production remains insufficiently explored. This study evaluated the effects of three LAB...

The Synergistic Interaction Between BAP and GA3 Eradicates Hyperhydricity and Enhances Biomass in Solanum tuberosum L. cv. ‘Superchola’ In Vitro Plantlets

The mass propagation of pre-basic seed potatoes (Solanum tuberosum L.) is frequently bottlenecked by the physiological degradation of in vitro plantlets, largely driven by cytokinin oversaturation. This study evaluated the morphogenic and physiological effects of the synergistic interaction between 6-Benzylaminopurine...
