This review systematically covers the molecular structure, classical functions, and complex regulatory networks of PIWIL4 in both cancerous and non-cancerous conditions and focuses on the molecular underpinnings of this duality—specifically, piRNA-guided targeting, differences in protein interaction networks, and the influence of cellular context.
Abstract
PIWIL4 is a core component of the PIWI/piRNA pathway. Its roles in germ cell development, transposon silencing, and genome stability maintenance are well established. Recent studies have shown that PIWIL4 is abnormally expressed in many cancers. There, it helps regulate key malignant processes—including cell cycle control, apoptosis, epithelial-mesenchymal transition, stemness maintenance, and metabolic reprogramming. However, its effects vary widely, it can promote or suppress tumours depending on the cancer type, and even across subtypes of the same cancer. This striking functional duality is highly context-dependent. This review systematically covers the molecular structure, classical functions, and complex regulatory networks of PIWIL4 in both cancerous and non-cancerous conditions. We focus on the molecular underpinnings of this duality—specifically, piRNA-guided targeting, differences in protein interaction networks, and the influence of cellular context. We also assess its promise and hurdles as a diagnostic biomarker and therapeutic target. A deeper understanding of these context-dependent mechanisms should inform the design of targeted interventions in precision oncology.
This review systematically synthesizes advances in the pivotal, context-dependent roles of BTG2 in non-tumor pathologies, including fibrotic, neurological, cardiovascular, inflammatory, metabolic and other systemic diseases.

Cellular heterogeneity in pancreatic ductal adenocarcinoma (PDAC) poses significant challenges for disease prognosis and limits the effectiveness of therapeutic strategies implemented in recent multicenter subtype-based clinical trials worldwide. A hallmark of PDAC is extensive intratumoral heterogeneity, which is la...

Non-coding RNAs serve as key regulators of gene expression at both the transcriptional and post-transcriptional levels. Among them, piRNAs, a class of small non-coding RNAs, have emerged as significant mediators in cancer biology. PIWI proteins, members of the Argonaute family highly expressed in germline cells, intera...

Glioblastoma multiforme (GBM) is an extremely virulent and treatment-resistant primary brain tumor that has a very rapid course, is commonly recurrent, and has very poor patient prognosis. Discovery of strong molecular targets is still urgent to further precision oncology in GBM. An integrative bioinformatics and compu...
