Integrated Single-Cell and Transcriptomic Analysis Suggests a Cuproptosis-Related Phenotype and Potential Therapeutic Target in Acute Pancreatitis

Acute pancreatitis (AP) is an acute inflammatory disorder of the exocrine pancreas with limited therapies. Whether cuproptosis, a copper-dependent cell death pathway, contributes to AP pathogenesis remains unknown. We integrated single-cell transcriptomics (GSE279876), single-cell transcriptomics (GSE188819), and human...

Integrative multi-omics analysis prioritizes compartment-specific candidate targets of gut microbiota metabolites in diabetic kidney disease

A compartment-specific “M-S-M-T” regulatory network in the DKD glomerulus and tubulointerstitium is proposed and will require validation in real-world microbiome, metabolomic and in vivo studies before any therapeutic inference can be drawn.

Context-dependent variant interpretation from Mendelian disease to genetic predisposition: a proof-of-concept using LPL

A context-dependent framework is proposed in which biallelic complete- or near-complete loss-of-function genotypes are interpreted as causative for FCS, whereas heterozygous variants are interpreted as predisposing to severe hypertriglyceridemia while retaining recognition of FCS carrier status.
