pH ‐Responsive Loureirin A‐Modified Sodium Alginate‐Based Hydrogel for Accelerating Skin Wound Healing

Wound repair demands multifunctional dressings that simultaneously provide antibacterial protection, promote angiogenesis, and enable responsive drug release. Loureirin A (LA), the principal active component of Dragon's Blood, exhibits antibacterial and pro‐healing effects; however, its poor water solubility and low local bioavailability limit therapeutic efficacy. Copper ions (Cu 2+ ) possess antibacterial activity but carry burst‐release toxicity risks. This study proposed a natural product–inorganic ion synergistic controlled‐release strategy: mesoporous polydopamine nanoparticles (MPDA) were employed to achieve high LA loading, and Cu 2+ was ionically crosslinked with sodium alginate (SA) to construct a pH‐responsive MCLS hydrogel, with the network stabilized by hydrogen and disulfide bonds. This system enabled coordinated LA and Cu 2+ release under weakly acidic wound conditions. The MCLS hydrogel showed a porous microstructure, appropriate mechanical properties, and pH‐dependent release. In vitro, it exhibited antibacterial activity against Staphylococcus aureus and Escherichia coli and demonstrated cytocompatibility. In a rat full‐thickness skin defect model, the hydrogel accelerated wound closure, promoted fibroblast migration and tube formation, and upregulated CD31 and VEGF‐A expression, indicating enhanced angiogenesis. This study provides an experimental basis for functional wound dressings based on the synergistic combination of natural active ingredients and inorganic ions.
