Nanoemulsion adjuvanted COBRA HA vaccine enhances protective antibody titers against H1N1 and H3N2 influenza viruses

ABSTRACT Adjuvants can enhance the elicited immune responses to recombinant hemagglutinin (HA) proteins following intramuscular vaccination. In this study, computationally optimized broadly reactive antigen (COBRA) designed HA proteins were adjuvanted with the W805EC oil-in-water nanoemulsion (“NE01”) and used to vacci...

Intranasal delivery of a COBRA-based protein vaccine adjuvanted with c-di-AMP enhances breadth of protective immune responses against seasonal influenza viruses

Seasonal influenza vaccines provide variable protection against antigenically drifted influenza A (H1N1 and H3N2) and influenza B viruses. Broadly protective approaches using computationally optimized broadly reactive antigens (COBRAs) can enhance immunity against diverse strains. Bis-(3′,5′)-cyclic dimeric adenosine m...

Computationally Optimized H1 and H3 Hemagglutinin Messenger RNA Vaccines Confer Broad Protective Immunity Against Modern Influenza Viruses

These findings demonstrate that COBRA HA encoding mRNA vaccines can effectively leverage immunological memory while expanding responses to conserved HA epitopes, supporting improved protection against antigenically drifted strains and represents a promising strategy for next-generation influenza vaccination.
