11 Long-Term Follow-Up from the OMNIVORE Trial: Response-Adaptive Nivolumab and Ipilimumab in Advanced Renal Cell Carcinoma (NCT03652142)

Abstract Background Immune checkpoint inhibitor-based combinations represent the standard of care for advanced renal cell carcinoma (RCC). The OMNIVORE trial (NCT03652142) investigated a response-adaptive strategy including treatment discontinuation in early nivolumab responders (Arm A) and salvage ipilimumab addition...

70 Lymphocyte Hsp70 is a Novel Predictive Biomarker for Response to Immune Checkpoint Inhibitors in Metastatic Renal Cell Carcinoma

A scRNA-seq atlas of tumors from metastatic RCC found a novel factor, Hsp70, that in lymphocytes strongly predicts response and greatly extended PFS after ICI, indicating that Hsp70 expression in lymphocytes predicts response to ICI across timepoints and cohorts.

10 Outcomes of Patients with Renal Cell Carcinoma Achieving Pathologic Complete or Major Response After Consolidative Nephrectomy Following Immune Checkpoint Inhibitor Therapy

Abstract Background Select patients with locally advanced or metastatic renal cell carcinoma (RCC) are candidates for consolidative nephrectomy (CN) following immune checkpoint blockade (ICB). Existing cohorts have been limited by heterogeneous pathological assessment and small sample sizes, limiting validation of path...

22 Phase II Study of Cemiplimab ± Fianlimab Following SBRT for Oligometastatic Clear-Cell RCC (LAG-BOOST) (NCT07223541)

Abstract Background Oligometastatic clear-cell renal cell carcinoma (ccRCC) is a biologically distinct, often indolent state where metastasis-directed therapy, including metastasectomy or SBRT, provides durable local control and delays systemic therapy. Randomized prospective trial evidence demonstrates a survival bene...

19 Dissecting ARNT Expression Patterns in RCC and Association with Outcomes

Abstract Background Hypoxia signaling is a key driver of renal cell carcinoma (RCC) and is mediated by HIF-dependent transcription. ARNT (HIF-1β) is an essential HIF binding partner, but its expression and clinical relevance in RCC are poorly defined. With ARNT inhibitors in development, characterizing ARNT expression...

8 Single Cell Transcriptomic Investigation of Renal Cell Carcinoma (RCC) Reveals Tissue Resident Memory Exhausted CD8+ T Cell Signature Associated with Resistance to Immune Checkpoint Inhibition (ICI)

A tissue residency gene program in Tex cells associated with non-response to immunotherapy was identified and nominates resident memory exhausted CD8+ T cells as a targetable subset of cells to improve CD8+ T cell-mediated anti-tumor immunity.

51 Discovery of Exhausted CD8+ T-cell States Associated with Clinical Outcomes in Renal Cell Carcinoma

It is found that baseline tumor infiltration by exhausted T cells with a resident memory phenotype (Tex-rm) is significantly associated with worse PFS in advanced RCC patients receiving ICI monotherapy or ICI+TKI combination therapy.

23 PRISM: Phase 1b of Retifanlimab and Ruxolitinib in Solid Malignancies Progressing on Prior Checkpoint Inhibition (NCT07219576)

The vision is to establish JAK inhibition as a tractable strategy for restoring checkpoint sensitivity across solid tumors, providing the foundation for a biomarker-driven randomized phase 2 trial in checkpoint-refractory disease.
