Receptor allostery promotes context-specific Sonic Hedgehog signaling during embryonic development

The authors identify a binding site on the Sonic Hedgehog signaling protein Smoothened for the allosteric regulator arachidonic acid (AA) and show that its disruption attenuates SHH- and sterol-stimulated SMO induction.

Developmental mis-specification of cardiac conduction and structure transcriptome in Brugada syndrome

The findings support a model in which non-SCN5A BrS is associated with early dysregulation of developmental transcriptional networks controlling cardiac conduction and extracellular matrix organization, whereas SCN5A-mediated BrS primarily results from sodium channel deficiency with limited developmental remodeling.
