Microglial dynamics associate with the spatial pattern of tau deposition in early Alzheimer’s disease

Neuroinflammation is implicated in Alzheimer’s disease (AD) pathogenesis, yet how changes in TSPO PET signal are associated with tau deposition, and how brain network architecture shapes this relationship remain poorly understood. We conducted a prospective, multimodal neuroimaging study in 56 participants: 35 amyloid-...

Network‐dependent remote effects and local tau burden jointly shape neuronal dysfunction in Alzheimer's disease

Abstract BACKGROUND Tau pathology in Alzheimer's disease (AD) propagates along the brain's connectome, yet the mechanisms by which tau drives neuronal dysfunction remain unclear. We hypothesized that dysfunction primarily emerges in regions close to tau‐harboring epicenters. METHODS One hundred thirty‐one amyloid‐posit...

Different functional connectivity gradients reflect aging and Alzheimer's disease.

It is shown that functional connectivity changes associated with aging and AD are strongly aligned with separate fundamental axes of hierarchical brain communication, which suggest that both AD and aging alter major but orthogonal functional pathways in the brain.

Activity-dependent epidemic spreading on multiscale brain networks predicts Alzheimer's disease progression

The results connect epidemic theory to neurodegeneration, implicate neuronal activity as a driver of Alzheimer's disease progression, and motivate activity-modulating therapies to slow or prevent pathological spread are connected.

Dynamic [18F]PI-2620 recording facilitates prediction of β-amyloid positivity and simultaneous staging of tau and neurodegeneration

Dynamic [18F]PI-2620 PET imaging facilitates assessment of the β-amyloid status and regional staging of tau and neurodegeneration during a single acquisition, and may reduce radiation exposure, streamline diagnostic workflows, and facilitate personalized disease profiling.

Mesoscale medial temporal lobe connectivity patterns relate to tau pathology and memory in older adults

Differential associations of age and AD pathology with mesoscale MTL-connectivity patterns show differential associations of age and AD pathology with mesoscale MTL-connectivity patterns, which may support memory function in the short term while contributing to subsequent memory decline.

Concordance Between [18F]Flortaucipir PET Visual Reads and CenTauR-Based Quantification.

Generalized additive models examined clinical progression on the basis of visual and CenTauRz-based assessments of tau PET positivity and highlighted the need for new quantification approaches to better capture the complex patterns of tau deposition, important for early AD detection and monitoring.
