This preclinical protocol presents an advanced computational and experimental framework for directed cancer treatment based on the mathematical modeling of p53-MDM2 cellular network dynamics. Instead of relying on conventional dose-dependent cytotoxicity, the approach finely tunes the degradation rate of MDM2 to push oncogenic networks into a self-sustaining oscillatory regime, ultimately triggering irreversible mitochondrial apoptosis. To bypass the physical diffusion limitations inherent in solid tumors, the protocol deploys a "Trojan Horse" active transport strategy utilizing engineered tumor-associated macrophages (TAMs).
