Targeting PRDX1-G6PD Interaction Promotes a Metabolic Vulnerability and Redox Imbalance in Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype with limited targeted options, and its progression is strongly driven by metabolic reprogramming and redox homeostasis. Here, we found that peroxiredoxin-1 (PRDX1) promotes TNBC progression by directly interacting with glucose-6-phosphate dehyd...

Blocking the m6Am methyltransferase PCIF1 in macrophages attenuates colitis progression by preventing neuronal damage

It is demonstrated that colonic macrophage-mediated extracellular matrix (ECM) remodeling orchestrates enteric neuronal maturation and dictates colitis progression and the m6Am machinery is illuminated as a promising therapeutic target for preventing neuronal loss in IBD patients.

PGK1-driven histone lactylation fuels MDSC-driven immune evasion and tumor progression in triple-negative breast cancer.

Functional assays demonstrated that PGK1-driven glycolysis in TNBC cells promotes lactate accumulation and H3K18la, which subsequently induce transcriptional activation of CCL5 and recruitment of MDSCs, thereby impairing CD8+ T cell function and fostering an immunosuppressive tumor microenvironment that facilitates imm...

Blocking the m6Am methyltransferase PCIF1 releases STAT1-mediated Th1 immunity to potentiate cancer immunotherapy

It is shown, by genomic deletion of PCIF1 specifically in T cells, that PCIF1 represses STAT1 translation via this important mRNA modification, and thereby preventing T cell activation and enforcing a naïve T cell phenotype.
