Neuroscientific foundations of meditation: an advanced analysis of neurochemical systems, neural network dynamics, and central functional plasticity

Background: The practice of meditation and mindfulness techniques has garnered significant interest within the international scientific community due to its potential to induce functional and structural plasticity in the Central Nervous System (CNS). This advanced narrative review aims to analyze, refine, and systematize the neuroscientific foundations of meditation, correlating ultrastructural synaptic physiology, complex neuroactive substance taxonomy, and specific neural pathway mapping with dynamic neuroimaging alterations and the core pillars of psychophysical practices. Methods: A comprehensive review of literature was conducted to evaluate the behavior and regulation of crucial neurochemical systems, including glutamatergic, gabaergic, dopaminergic, serotonergic, noradrenergic, and cholinergic networks. Results: Functional neuroimaging evidence demonstrates a significant metabolic reorganization in the meditating brain compared to the baseline state, characterized by the attenuation of the Default Mode Network (DMN) and the strengthening of the Central Executive Network (CEN). Furthermore, down-regulation of the hyperactive amygdalofugal alarm circuits yields measurable anxiolytic phenotypes. Conclusion: Meditation acts as a potent modulator of neurochemical homeostasis, executive attention, and the attenuation of stress- and anxiety-related pathways, scientifically validating its integration into evidence-based clinical and academic settings.
