Regulation of mRNA poly(A) tail length governs mammalian body plan formation in gastruloids

A single-cell CRISPR screening platform is established in gastruloids to identify Cnot8-dependent mRNA deadenylation as a driver of mesoderm patterning and identify the deadenylase Cnot8 as a fundamental mechanism linking cellular identity with morphogenetic signaling during mammalian body plan formation.

Mapping the NFYA 3′UTR landscape identifies alternative polyadenylation as a targetable vulnerability in prostate cancer

Findings establish NFYA  3′UTR lengthening in PCa cells via CRISPR/Cas9 or ASOs diminished NF-YA protein levels and suppressed aggressive tumor traits both in vitro and in vivo, and provide proof-of-concept for gene-specific, APA-directed therapeutic strategies.
