Beyond amyloid: Contributions of neuroinflammation, cerebrovascular disease, and neurodegeneration to memory impairment in the oldest‐old, the LifeAfter90 Study

Neuroinflammatory processes may play a central role in promoting mixed pathologies and cognitive impairment in the oldest‐old. We hypothesize that elevated neuroinflammation, indicated by reactive astrocytosis, acts on cerebrovascular disease and separately, on amyloid beta (Aβ) pathology, to promote downstream medial temporal lobe atrophy and memory impairment.

Amyloid PET Quantitation and Centiloid Thresholds in the Diagnosis of Alzheimer Disease: An Individual Participant Data Meta-Analysis.

A double-cutoff analysis suggest that scans in the 11 to 26 Centiloid range should be interpreted with caution depending on the context of use, and positivity cutoffs converged around 18 Centiloids (data-driven) and 27 Centiloids (visual reads).

Parallel neuroinflammatory pathways to cerebrovascular burden and amyloid beta in Alzheimer's disease

Higher plasma YKL‐40 was associated with greater white matter hyperintensity (WMH), whereas higher plasma GFAP was related to increased 18F‐florbetapir (FBP) standardized uptake value ratio (SUVR) and higher plasma p‐tau217 was associated with reduced MTL cortical thickness and hippocampal volume.
